Recombinant Antibody Expression

To mimic the behavior of a recombinant human IgG antibody (targeting a human protein) in humans the best, specificity-matching recombinant mouse IgG antibodies derived from phage display mouse scFv / Fab libraries (targeting the mouse protein) are frequently produced and used in animal models. We use a modified vector to produce recombinant antibodies in HEK293 or CHO cells for transient or stable expression. Recombinant Antibody Fragment Expression

scFv

Compared with mAbs, scFv’s have a small size and offer several advantages, including low kidney uptake, rapid blood clearance, greater tumor penetration, and less negative response from the human immune system. scFv fragments could be expressed in both bacterial cells and mammalian cells. Our service provides scFv construction via two different methods, i.e., scFv produced from synthetic DNA or via accession number.

Fab / Fabʹ

Fab fragments differ from scFv’s as they are composed of one constant and one variable domain of each of the heavy and the light chains. Fab fragments can be created using papain, which could cleave an immunoglobulin monomer into two Fab fragments and an Fc fragment. If the variable domains had been sequenced, the fragments could be produced by recombinant expression.

sdAb / VHH

A single-domain antibody (sdAb, VHH, or nanobody) is a type of antibody fragments consisting of a single monomeric variable antibody domain. With a molecular weight of only 12–15 kDa, single-domain antibodies are much smaller than common antibodies (150–160 kDa), and even smaller than scFv fragments (25 kDa): offer some advantages, including outstanding penetrability and high stability, and can be expressed in both eukaryotic and prokaryotic systems.

Fc Fusion Proteins

Fc fusion proteins are composed of an immunoglobulin Fc domain that is directly linked to another peptide or protein of interest. Fc fusion proteins can play a role in dimerization, in vivo half-life, and receptor activation and affinity for the cognate ligand.

Advantages of Recombinant Antibody

Traditional antibodies need animal immunization. Different batches of animals may generate different antibodies, which can be caused by many factors, such as individual animal health, immune status, and efficiency of immunization program. Importantly, animal-derived antibodies often cause immune reactions in patients, limiting their use as clinical therapies. Unlike traditional antibodies, recombinant antibodies offer several advantages over both traditional monoclonal and polyclonal antibodies in the field of human medicine and research.


  • Animal-free: Recombinant antibodies have the identical DNA and protein sequence, in vitro production can be implemented. This process does not require use of animals, eliminating animal discomfort or pain.
  • Decreased production time: Using protein expression system, such as mammalian system, the production time of recombinant can be weeks compared to months for monoclonal antibodies.
  • Consistency and reproducibility: Recombinant antibodies are constructed by isolation of the respective antibody genes, which means the production process is controlled and reliable. Meanwhile, this leads to antibodies with very little batch-to-batch variability, giving highly reproducible results.
  • Reformatting: Unlike hybridoma-derived antibodies, recombinant antibodies can be easily converted into different species, different isotypes or subtypes, and different formats.
