Compared with mAbs, scFv’s has a small size and offer several advantages, including low kidney uptake, rapid blood clearance, greater tumor penetration, and less negative response from the human immune system. scFv fragments could be expressed in both bacterial cells and mammalian cells. Our service provides scFv construction via two different methods, i.e., scFv produced from synthetic DNA or via accession number.
Fab fragments differ from scFv’s as they are composed of one constant and one variable domain of each of the heavy and the light chains. Fab fragments can be created using papain, which could cleave an immunoglobulin monomer into two Fab fragments and an Fc fragment. If the variable domains had been sequenced, the fragments could be produced by recombinant expression.
A single-domain antibody (sdAb, VHH, or nanobody) is a type of antibody fragments consisting of a single monomeric variable antibody domain. With a molecular weight of only 12–15 kDa, single-domain antibodies are much smaller than common antibodies (150–160 kDa), and even smaller than scFv fragments (25 kDa): offer some advantages, including outstanding penetrability and high stability, and can be expressed in both eukaryotic and prokaryotic systems.
Fc fusion proteins are composed of an immunoglobulin Fc domain that is directly linked to another peptide or protein of interest. Fc fusion proteins can play a role in dimerization, in vivo half-life, and receptor activation and affinity for the cognate ligand.
